siRNA targeting CD44 is a target-specific RNA interference (RNAi) modality designed to silence the expression of CD44, a cell-surface glycoprotein overexpressed in various cancers and cancer stem cells. CD44 plays a critical role in cell adhesion, migration, proliferation, and epithelial-to-mesenchymal transition (EMT), and is associated with therapeutic resistance and metastasis. By delivering small interfering RNA (siRNA) sequences complementary to CD44 mRNA, this approach triggers the RNA-induced silencing complex (RISC) to degrade CD44 transcripts, thereby downregulating CD44 protein expression. While widely utilized as a research reagent to study CD44 biology and overcome drug resistance in preclinical models, various delivery systems, such as nanoparticles, exosomes, and peptide carriers, are being investigated to translate CD44-targeted siRNA therapies into clinical applications.
