siRNA targeting CD45 is a therapeutic modality that utilizes small interfering RNA (siRNA) to silence the expression of the PTPRC gene, which encodes the CD45 protein. CD45, also known as the common leukocyte antigen, is a transmembrane protein tyrosine phosphatase expressed on all nucleated hematopoietic cells and is essential for T-cell and B-cell receptor-mediated activation. By employing RNA interference (RNAi), this approach aims to reduce CD45 levels, thereby modulating immune responses. Preclinical research has explored the use of CD45 siRNA delivered via lipid nanoparticles (LNPs) or other vectors for applications such as treating autoimmune diseases, preventing graft-versus-host disease (GvHD), and managing hematologic malignancies like leukemia. While currently primarily used as a research tool, it represents a targeted strategy for selective immunosuppression and immune system modulation.
