siRNA targeting KIFC1 is a therapeutic modality designed to silence the expression of Kinesin Family Member C1 (KIFC1), also known as HSET. KIFC1 is a minus-end-directed microtubule motor protein that is non-essential for normal cells but critical for cancer cells with supernumerary centrosomes. These cancer cells rely on KIFC1 to cluster extra centrosomes into two poles during mitosis, avoiding lethal multipolar divisions. By utilizing small interfering RNA (siRNA) to degrade KIFC1 mRNA, the protein's function is inhibited, leading to centrosome declustering, multipolar spindle formation, anaphase catastrophe, and apoptosis. This approach is being investigated as a novel therapeutic strategy for cancers characterized by centrosome amplification and poor prognosis, such as small cell lung cancer (SCLC).
