Small interfering RNA targeting LASS2 is an RNA interference (RNAi) agent designed to silence the expression of the Longevity Assurance Homolog 2 (LASS2) gene, also known as Ceramide Synthase 2 (CERS2). LASS2 is a recognized tumor suppressor that inhibits cancer metastasis by binding to the C subunit of vacuolar H+-ATPase (V-ATPase), thereby regulating its activity. In experimental models, particularly bladder cancer, the application of LASS2-specific siRNA leads to the downregulation of LASS2 protein levels, which results in increased V-ATPase activity and a subsequent rise in extracellular hydrogen ion concentration. This acidification triggers the activation of secreted matrix metalloproteinases, specifically MMP-2 and MMP-9, which facilitates tumor cell proliferation, invasion, and survival. Furthermore, silencing LASS2 has been shown to suppress apoptosis induced by chemotherapeutic agents such as doxorubicin. While primarily utilized as a research tool to investigate the molecular mechanisms of tumor progression and ceramide metabolism, it represents a specific application of RNA therapy in oncology research.
