siRNA targeting MALT1 refers to a class of RNA interference (RNAi) therapeutics designed to silence the expression of the Mucosa-Associated Lymphoid Tissue Lymphoma Translocation Protein 1 (MALT1). MALT1 is a unique paracaspase that plays a dual role as a scaffold and a protease within the CBM (CARMA1-BCL10-MALT1) complex, which is essential for the activation of the NF-κB signaling pathway following antigen receptor stimulation in B and T cells. Constitutive MALT1 activity is a hallmark of several lymphoid malignancies, including Activated B-Cell-like Diffuse Large B-Cell Lymphoma (ABC-DLBCL) and MALT lymphoma. By utilizing the RNA-induced silencing complex (RISC), these siRNAs facilitate the sequence-specific degradation of MALT1 mRNA, thereby reducing MALT1 protein levels and inhibiting downstream oncogenic signaling and pro-inflammatory cytokine production. While several small molecule MALT1 inhibitors are currently in clinical development, siRNA-based approaches are primarily utilized in preclinical research to validate MALT1 as a therapeutic target and to study its role in immune cell signaling and lymphomagenesis.
