siRNA targeting PD-L1 is a therapeutic modality designed to silence the expression of Programmed Death-Ligand 1 (PD-L1) through the RNA interference (RNAi) pathway. By delivering small interfering RNA sequences specific to the PD-L1 mRNA, this approach leads to the sequence-specific degradation of the transcript, thereby reducing the protein levels of PD-L1 on the surface of tumor cells and other cells within the tumor microenvironment. PD-L1 is a key immune checkpoint molecule that, when bound to PD-1 on T cells, inhibits T-cell activation and promotes immune evasion. Silencing PD-L1 restores the anti-tumor activity of cytotoxic T lymphocytes and natural killer cells. Research focuses on various delivery vehicles, such as lipid nanoparticles (LNPs), polymeric nanocapsules, and attenuated bacterial vectors, to overcome the challenges of siRNA stability and targeted delivery.
