siRNA targeting PLXND1 is an experimental RNA interference (RNAi) therapeutic approach designed to silence the expression of the PlexinD1 (PLXND1) receptor. PLXND1 is a semaphorin receptor that plays a critical role in cytoskeletal organization and cell adhesion. Research indicates that PLXND1 is significantly upregulated in neuroendocrine prostate cancer (NEPC), a highly aggressive and therapy-resistant form of prostate cancer that often arises through lineage plasticity. By utilizing siRNA to degrade PLXND1 mRNA, researchers have demonstrated reduced proliferation in NEPC cell lines and decreased viability in patient-derived xenograft (PDX) organoids. This suggests that targeting the PLXND1 pathway may provide a novel strategy for treating advanced prostate cancer and overcoming resistance to androgen receptor signaling inhibitors.
