siRNA targeting PSMC6 is an experimental RNA interference (RNAi) therapeutic designed to silence the expression of the PSMC6 gene, which encodes the 26S proteasome regulatory subunit 6B. PSMC6 was identified through CRISPR/Cas9 dropout screening as a critical survival factor for cisplatin-resistant ovarian carcinoma cells. By utilizing small interfering RNAs (siRNAs) to degrade PSMC6 mRNA, researchers have demonstrated efficient down-regulation of the target at both the mRNA and protein levels. This knockdown results in reduced cancer cell clonogenicity in both 2D and soft agar assays and is associated with the down-regulation of the canonical MAPK signaling pathway. Furthermore, clinical data indicates that PSMC6 expression levels correlate with tumor stage in ovarian carcinoma, suggesting its potential as a therapeutic target for aggressive or drug-resistant disease.
