Overview

siRNA targeting PSMD6 is an experimental RNA interference (RNAi) therapeutic approach designed to treat multiple myeloma by silencing the expression of the PSMD6 gene. PSMD6 encodes a non-ATPase subunit of the 19S regulatory particle of the 26S proteasome, which is essential for protein homeostasis. In multiple myeloma, which is highly dependent on proteasome function, knockdown of PSMD6 leads to the accumulation of K48-linked polyubiquitinated proteins, induction of endoplasmic reticulum (ER) stress, G0-G1 cell cycle arrest, and apoptosis. This strategy aims to overcome resistance to traditional 20S proteasome inhibitors by targeting the regulatory particle upstream. Research is primarily being conducted at the Dana-Farber Cancer Institute.
