siRNA targeting SATB2 refers to small interfering RNA molecules designed to silence the expression of the Special AT-rich sequence-binding protein 2 (SATB2). SATB2 is a transcription factor and chromatin remodeler that regulates gene expression in a tissue-specific manner. In the context of colorectal cancer (CRC), SATB2 is often lost during the progression from adenoma to carcinoma, and its loss is associated with an aggressive phenotype, chromosomal instability, and a pro-inflammatory immune microenvironment. Research using SATB2-targeting siRNA in cell lines like SW480 has demonstrated that reducing SATB2 levels promotes epithelial-mesenchymal transition and increases the expression of checkpoint genes like PD-1 and CTLA-4. While currently utilized primarily as a research tool to study tumor suppression and immune regulation, SATB2 modulation represents a potential area of interest in oncology and inflammatory bowel disease.
