The combination of sirolimus and vandetanib has been studied primarily for the treatment of recurrent glioblastoma. Sirolimus (also known as rapamycin) is an FDA-approved drug that inhibits mammalian target of rapamycin (mTOR), a member of the phosphoinositide 3-Kinase signaling pathway. Vandetanib is a drug that inhibits epidermal growth factor receptor (EGFR) and vascular endothelial growth factor receptor 2 (VEGFR2). The combination of these two drugs targets specific molecular alterations in glioblastoma with the aim of more effectively killing tumor cells and potentially increasing survival.
