Sirolimus (Rapamune) acts by inhibiting the mammalian target of rapamycin (mTOR) to block T-cell proliferation, while mycophenolate mofetil (CellCept) inhibits inosine monophosphate dehydrogenase (IMPDH) to suppress lymphocyte purine synthesis.
