Overview

SIRPα antisense morpholino is a synthetic antisense oligonucleotide designed to knockdown the expression of Signal Regulatory Protein Alpha (SIRPα). While SIRPα is traditionally known for its role on myeloid cells where it binds CD47 to inhibit phagocytosis, research indicates it is also expressed on malignant cells in glioblastoma (GBM). This morpholino-mediated blockade inhibits mitochondrial fatty acid β-oxidation (FAO) in GBM cells, leading to decreased mitochondrial ATP production. Furthermore, targeting SIRPα on GBM cells enhances microglia-mediated cytotoxicity, suggesting that SIRPα regulates both tumor metabolism and immune evasion in the brain tumor microenvironment.
