SIRT6-mRNA.019 is a preclinical mRNA-based therapeutic candidate developed by SirtLab for the treatment of osteoarthritis. It utilizes lipid nanoparticle (LNP) technology to deliver mRNA encoding the SIRT6 (Sirtuin 6) protein directly to affected joint tissues. SIRT6 is a NAD+-dependent deacetylase and mono-ADP-ribosyltransferase that plays a critical role in maintaining joint homeostasis by regulating DNA repair, telomere maintenance, and inflammatory signaling. By restoring or enhancing SIRT6 levels in chondrocytes, the therapy aims to inhibit cartilage degradation and reduce the progression of osteoarthritis. The program has demonstrated proof-of-concept in animal models.
