Overview

Sirtratumab vedotin (AGS15E, ASG-15ME) is an investigational antibody-drug conjugate (ADC) designed for the treatment of metastatic urothelial carcinoma. It consists of a monoclonal antibody targeting SLIT and NTRK-like protein 6 (SLITRK6), a neuronal transmembrane protein highly expressed in urothelial carcinoma cells but with limited expression in normal tissues. The antibody is conjugated via a protease-cleavable linker to monomethyl auristatin E (MMAE), a potent microtubule-disrupting cytotoxic agent. Upon binding to SLITRK6 on tumor cells, the ADC is internalized and MMAE is released intracellularly, leading to disruption of tubulin polymerization and subsequent apoptosis. Sirtratumab vedotin was developed to address unmet needs in patients with advanced or metastatic urothelial cancer who have failed prior therapies[1][2][3][5][6].
