siSept9 is a small interfering RNA (siRNA) designed to specifically silence the expression of Septin-9 (Sept9), a member of the septin family of GTP-binding proteins that act as a component of the cytoskeleton and scaffolding proteins for various cellular functions. In preclinical research, siSept9 is used to investigate the role of Septin-9 in maintaining the structural integrity of desmosomal cell-cell junctions. Studies in squamous cell carcinoma (SCC9) and breast cancer (T47D) cell lines have shown that siSept9-mediated knockdown of Septin-9 leads to the significant disruption of desmosomal junctions and the mislocalization of key desmosomal proteins, such as Desmoglein-2 (Dsg2) and Plakophilin-2 (PKP2), without affecting their total protein levels. This disruption of cellular adhesion is a critical mechanism that may contribute to enhanced cancer motility, invasion, and metastatic potential during tumor progression.
