Overview

siSIRT3 is a small interfering RNA (siRNA) designed to silence the expression of Sirtuin 3 (SIRT3), a mitochondrial NAD+-dependent deacetylase. SIRT3 is a member of the sirtuin family (Class III histone deacetylases) and is primarily localized in the mitochondria, where it regulates metabolic processes, oxidative stress, and cell survival. In the context of oncology, particularly melanoma, SIRT3 has been identified as having pro-proliferative functions. siSIRT3 is utilized in preclinical research to evaluate the therapeutic potential of SIRT3 inhibition in models such as BrafV600E/PtenNULL mice and patient-derived xenografts (PDX). Research findings suggest that while siSIRT3 can reduce tumor growth and metastatic lung nodules, its efficacy as a monotherapy may be limited, indicating that the concomitant inhibition of multiple sirtuins (such as SIRT1 and SIRT3) might be a more effective strategy for melanoma management.
