siSLAMF8 is a small interfering RNA (siRNA) designed to specifically inhibit the expression of Signaling Lymphocytic Activation Molecule Family Member 8 (SLAMF8). SLAMF8 is predominantly expressed in macrophages and plays a critical role in the tumor immune microenvironment by promoting the polarization of tumor-associated macrophages (TAMs) toward the immunosuppressive M2 phenotype. By silencing SLAMF8, siSLAMF8 facilitates the repolarization of TAMs toward an anti-tumor (CD86+) phenotype, reduces the infiltration of exhausted CD8+ T cells, and enhances the activity of cytotoxic CD8+ T cells. Preclinical studies in colorectal cancer models suggest that siSLAMF8 can delay tumor progression and sensitize tumors to immune checkpoint inhibitors, such as anti-PD-1 therapy, by modulating the PI3K/AKT and JAK/STAT3 signaling pathways.
