SIST2301

SIST2301 is a small molecule potassium-competitive acid blocker (P-CAB) currently under development by Samik Pharmaceutical. It is being developed as a generic version of tegoprazan (originally developed by HK inno.N as K-CAB). As a P-CAB, SIST2301 functions by competitively inhibiting the potassium-binding site of the gastric H+/K+-ATPase enzyme, which is the final step of gastric acid secretion. This mechanism allows for rapid, potent, and reversible inhibition of acid production without the need for acid activation required by traditional proton pump inhibitors (PPIs). Samik Pharmaceutical is positioning SIST2301 as a "first generic" candidate in the South Korean market, targeting indications such as gastroesophageal reflux disease (GERD). The drug is currently in Phase 1 clinical trials to establish bioequivalence or comparative efficacy and safety.
