Overview

DKF-447 is a fixed-dose combination (FDC) therapy being developed by Dongkook Pharmaceutical for the treatment of type 2 diabetes mellitus. It combines two antihyperglycemic agents with complementary mechanisms of action: sitagliptin, a dipeptidyl peptidase-4 (DPP-4) inhibitor, and dapagliflozin, a sodium-glucose cotransporter 2 (SGLT2) inhibitor. Sitagliptin works by increasing the levels of active incretin hormones, which stimulates insulin secretion and reduces glucagon levels in a glucose-dependent manner. Dapagliflozin acts by inhibiting glucose reabsorption in the proximal tubule of the kidney, thereby promoting urinary glucose excretion. This combination is designed to provide synergistic glycemic control and improve patient compliance through a simplified dosing regimen.
