SIPS-2209 is a fixed-dose combination (FDC) drug candidate developed by Samik Pharmaceutical for the treatment of type 2 diabetes mellitus. It consists of two established antidiabetic agents: sitagliptin and pioglitazone. Sitagliptin is a dipeptidyl peptidase-4 (DPP-4) inhibitor that works by slowing the inactivation of incretin hormones, thereby increasing insulin secretion and decreasing glucagon levels in a glucose-dependent manner. Pioglitazone is a thiazolidinedione that acts as a selective agonist for the peroxisome proliferator-activated receptor gamma (PPAR-γ), which improves insulin sensitivity in muscle and adipose tissue and inhibits hepatic gluconeogenesis. By combining these two mechanisms, SIPS-2209 aims to provide comprehensive glycemic control by addressing both insulin resistance and impaired insulin secretion. The drug has been evaluated in Phase 1 clinical trials to establish bioequivalence between the fixed-dose combination and the co-administration of its individual components.
