Sitagliptin phosphate + metformin hydrochloride is a fixed-dose combination small molecule therapy developed by Saile Pharmaceutical Technology (Shanghai) Co., Ltd. for the management of type 2 diabetes mellitus. The product combines sitagliptin, a dipeptidyl peptidase-4 (DPP-4) inhibitor, with metformin hydrochloride, a biguanide. Sitagliptin works by slowing the inactivation of incretin hormones, thereby increasing insulin release and decreasing glucagon levels in a glucose-dependent manner. Metformin reduces hepatic glucose production, decreases intestinal absorption of glucose, and improves insulin sensitivity by activating AMP-activated protein kinase (AMPK), specifically involving subunits such as PRKAB1. This formulation is a controlled-release tablet (100 mg/1000 mg) designed for oral administration and has been evaluated in bioequivalence studies against the reference drug Janumet XR.
