Drug intelligence / Profile preview
siTLR3-R9 is a preclinical small interfering RNA (siRNA) nanoparticle formulation designed for the treatment of inflammatory bladder diseases, such as interstitial cystitis. Developed by researchers at West China Hospital of Sichuan University, the therapeutic consists of siRNA targeting Toll-like receptor 3 (TLR3) complexed with nona-arginine (R9), a cell-penetrating peptide, to form self-assembled nanoparticles. To prevent rapid clearance by urine, the nanoparticles are co-administered within a mucoadhesive hydrogel microsphere system (composed of polyvinyl alcohol, gelatin, citric acid, and tannic acid) that physically anchors the siRNA to the bladder wall. In preclinical models of cyclophosphamide-induced cystitis, intravesical instillation of this formulation successfully silenced TLR3 expression, leading to reduced bladder inflammation, downregulation of pro-inflammatory cytokines (TNF-alpha, IL-6), and restoration of the urothelial barrier.
