Drug intelligence / Profile preview
This combination regimen consists of sitneprotafib (JAB-3312), a potent and selective small-molecule allosteric inhibitor of Src homology 2 domain-containing protein tyrosine phosphatase 2 (SHP2), combined with the chemotherapy regimen VIT (vincristine, irinotecan, and temozolomide). Sitneprotafib is developed by Jacobio Pharmaceuticals (and licensed to Allist in China). The combination is being evaluated in a Phase Ib/II investigator-initiated clinical trial in the United States for the treatment of patients with relapsed or refractory malignant peripheral nerve sheath tumors (MPNST). SHP2 acts as a key node in the RTK/RAS/MAPK signaling pathway, and its inhibition prevents adaptive resistance and feedback reactivation of MAPK signaling. Vincristine is a microtubule inhibitor, irinotecan is a topoisomerase I inhibitor, and temozolomide is an alkylating agent. Together, this combination aims to synergistically inhibit tumor growth and overcome resistance in RAS-pathway-driven malignancies like MPNST.
