siTUBB2B-gold nanoparticles is a preclinical therapeutic candidate consisting of small interfering RNA (siRNA) targeting the TUBB2B gene, delivered via gold nanoparticles. TUBB2B is a β-tubulin isoform that has been identified as a driver of brain metastatic colonization in triple-negative breast cancer (TNBC). The drug works by silencing TUBB2B expression, which inhibits tumor growth and disrupts gap junction-mediated communication between TNBC cells and astrocytes in the brain niche. Developed by researchers at the City University of Hong Kong, this agent is being investigated for its potential to treat brain metastases, particularly when used in combination with Akt pathway inhibitors such as ipatasertib (GDC-0068) or capivasertib.
