Overview

Sivifene is a small-molecule antineoplastic agent and immunomodulator that was developed as a topical treatment for cutaneous cancer metastases. It was primarily investigated for high-grade squamous intraepithelial lesions (HSIL) associated with human papillomavirus (HPV) infection and invasive carcinomas of the anogenital area, including cervical, vaginal, and anal cancers. The drug reached phase II clinical trials before its development was discontinued. Although initially thought to be a selective estrogen receptor modulator due to structural similarity to tamoxifen, subsequent research showed that sivifene does not bind the estrogen receptor or possess antiestrogenic activity. Its mechanism of action remains unknown but is believed to involve immunomodulation through upregulation of the CD45 T lymphocyte cell surface receptor[1][3][5].
