SIWA318H is a proprietary humanized monoclonal antibody developed to selectively target an advanced glycation end product (AGE) biomarker found on dysfunctional, disease-causing cells. These include cells exhibiting abnormally high glycolysis (the Warburg effect) and oxidative stress—characteristics common to cancer cells, senescent cells, and some microbially infected cells. The antibody binds specifically to this AGE biomarker (notably Nε-carboxymethyl lysine or CML), leading to immune-mediated destruction of the targeted cell via mechanisms such as antibody-dependent cell-mediated cytotoxicity (ADCC), opsonization, and phagocytosis. Preclinical studies have demonstrated that SIWA318H can bind pancreatic cancer cells and cancer-associated fibroblasts, reduce tumor growth in mouse models of pancreatic ductal adenocarcinoma (PDAC), decrease the number of senescent and profibrotic cells in tumors, and increase rates of complete remission. The drug is being developed primarily for oncology indications but may also have potential applications in aging-related diseases due to its ability to clear senescent or damaged cells[1][3][4][5][6][7].
