Overview

siWee1 is a small interfering RNA (siRNA) designed to silence the expression of the Wee1 kinase, a key regulator of the G2/M cell cycle checkpoint. By inhibiting Wee1, siWee1 prevents the inhibitory phosphorylation of CDK1 (Tyr15), leading to premature entry into mitosis or cell cycle arrest in G1/S and S phases, particularly in cancer cells with dysfunctional G1 checkpoints. In preclinical studies of malignant melanoma, siWee1 treatment has been shown to induce DNA damage (marked by γ-H2AX), reduce cell viability, and trigger apoptosis through caspase-3 and PARP cleavage. It is primarily used as a research tool to validate Wee1 as a therapeutic target in various malignancies, including melanoma.
