siWnt5a is a small interfering RNA (siRNA) designed to target and silence the expression of Wnt family member 5A (Wnt5a), a non-canonical Wnt ligand. In advanced prostate cancer, particularly enzalutamide-resistant castration-resistant prostate cancer (CRPC), Wnt5a is often upregulated and contributes to neural lineage plasticity and treatment resistance. A specific bioengineered version, tRNA-siWnt5a, utilizes a tRNA scaffold to improve stability and delivery. By knocking down Wnt5a, the drug suppresses the non-canonical Wnt5a/FZD2 signaling pathway, inhibits tumor cell proliferation and migration, and resensitizes resistant cells to androgen receptor (AR) antagonists like enzalutamide.
