siXIAP is a small interfering RNA (siRNA) designed to silence the expression of the X-linked inhibitor of apoptosis protein (XIAP), also known as BIRC4. XIAP is a potent endogenous inhibitor of apoptosis that functions by binding to and inhibiting initiator caspase-9 and effector caspases-3 and -7. In many cancers, including glioblastoma, XIAP is overexpressed, contributing to tumor cell survival, invasion, and resistance to chemotherapy and radiotherapy. By targeting XIAP mRNA for degradation, siXIAP reduces the levels of the XIAP protein, thereby lowering the threshold for apoptosis and sensitizing cancer cells to cell death triggers. Research has demonstrated that siXIAP can induce TNF-alpha-mediated apoptosis, lead to the loss of mitochondrial membrane potential, and inhibit the proliferation of malignant glioma cells in both in vitro and xenograft models.
