Overview

SJ-815 is a genetically engineered oncolytic vaccinia virus (Western Reserve strain) developed for the treatment of solid tumors, including pancreatic cancer and melanoma. The virus is modified by the deletion of the thymidine kinase (TK) gene to enhance tumor-selective replication and the insertion of two human transgenes: interferon beta 1 (IFNB1) and carboxylesterase 2 (CES2). The IFNB1 transgene promotes local immune activation and exerts direct anti-proliferative effects, while the CES2 transgene encodes an enzyme that facilitates the intratumoral conversion of the prodrug irinotecan into its active, highly cytotoxic metabolite, SN-38. This multi-modal approach combines direct viral oncolysis with localized chemotherapy sensitization and immunotherapy. Preclinical studies have demonstrated that SJ-815, particularly when used in combination with irinotecan, generates significant tumor regression and prolongs survival in various cancer models.
