Overview

SJ3149 is a selective and potent molecular glue degrader of casein kinase 1 alpha (CK1α), developed by St. Jude Children's Research Hospital. It is a small molecule that functions by binding to CK1α and recruiting it to the cereblon (CRBN) E3 ubiquitin ligase complex. This recruitment facilitates the polyubiquitination of CK1α, leading to its subsequent degradation via the 26S proteasome. SJ3149 has demonstrated broad antiproliferative activity in various human cancer cell lines, showing particular potency against acute myeloid leukemia (AML), as well as other hematological malignancies and solid tumors. The compound is orally bioavailable and is currently utilized as a preclinical research tool to study the therapeutic potential of CK1α degradation in oncology.
