Overview

SJCARB7H3_41BBL is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by St. Jude Children's Research Hospital. It is specifically engineered to target B7-H3 (CD276), a transmembrane protein highly expressed in pediatric solid tumors, including approximately 80% of osteosarcomas. The CAR construct features a CD8α/CD28 backbone and is uniquely modified to express the 4-1BB ligand (41BBL) on the cell surface to provide sustained costimulatory signals, enhancing T-cell persistence and anti-tumor activity. It is currently being evaluated in Phase II clinical trials as a consolidative treatment for patients with newly diagnosed high-risk metastatic osteosarcoma following standard MAP (methotrexate, doxorubicin, and cisplatin) chemotherapy.
