Overview

SJP1901 is a small molecule inhibitor of the Hippo pathway, specifically designed to target TEA domain (TEAD) transcription factors by directly inhibiting their S-palmitoylation. Palmitoylation of cysteine residues within the TEAD protein is a critical post-translational modification required for its stability and its ability to form functional transcriptional complexes with YAP and TAZ co-activators. By disrupting this process, SJP1901 prevents the expression of oncogenic target genes such as CTGF and CYR61, which drive tumor growth and proliferation. Developed by Samjin Pharm, SJP1901 has demonstrated potent anti-tumor activity in preclinical models of malignant mesothelioma and other Hippo pathway-dependent cancers, while showing minimal toxicity in normal cell lines.
