SJYC07 Intermediate-Risk Therapy is a risk-adapted treatment regimen developed by St. Jude Children's Research Hospital for young children (typically under age 3 or 5) newly diagnosed with various central nervous system (CNS) tumors, including medulloblastoma, ependymoma, and high-grade glioma. The regimen is part of the SJYC07 phase 2 clinical trial, which aims to improve survival while minimizing neurocognitive toxicity by using intensive chemotherapy to delay or reduce the field of radiation therapy. The intermediate-risk protocol specifically involves four cycles of induction chemotherapy (high-dose methotrexate, vincristine, cisplatin, and cyclophosphamide), followed by consolidation with focal radiotherapy (or further chemotherapy to delay radiation in infants), and six cycles of maintenance chemotherapy (cyclophosphamide, topotecan, and either erlotinib or etoposide).
