**SK-01** is a novel fusion protein, termed a synerkine, combining interleukin-4 (IL-4) and interleukin-10 (IL-10) to simultaneously inhibit **IL-4Rα** (interleukin-4 receptor subunit alpha) and **IL-10R** (interleukin-10 receptor). Developed by Synerkine Pharma, a Dutch biopharmaceutical company, it targets chronic pain conditions driven by inflammatory cytokines, offering a non-opioid mechanism with improved side-effect profiles over existing analgesics. SK-01 has received **European Orphan Drug Designation** for complex regional pain syndrome (CRPS), a severe chronic pain disorder often triggered by trauma and linked to immune dysregulation, and is advancing toward first-in-human clinical trials with clinical-grade material in production.[1][2]
