Overview

SK-0348 is a transdermal patch formulation of **tamsulosin hydrochloride**, a selective alpha-1 adrenoceptor antagonist, developed by **Sanwa Kagaku Kenkyusho**. Tamsulosin is traditionally administered orally for the treatment of benign prostatic hyperplasia (BPH) to improve lower urinary tract symptoms by relaxing smooth muscle in the prostate and bladder neck. The transdermal formulation was designed to provide sustained drug release and potentially improve patient compliance or reduce systemic side effects associated with peak plasma concentrations. However, in a Phase I first-in-human, single-dose, dose-escalation study in healthy adult male volunteers (jRCT2071220001), while the patch was generally well-tolerated with no major safety concerns, a clear dose-response relationship could not be established due to low systemic plasma concentrations of the drug. Consequently, the development of this specific transdermal formulation appears to have been terminated or discontinued, as it is no longer listed in the company's active R&D pipeline.
