SK-1011 is an orally administered small molecule calpain inhibitor being developed by Sanwa Kagaku Kenkyusho for the treatment of age-related macular degeneration (AMD), specifically subfoveal choroidal neovascularization (CNV). Calpains are a family of calcium-dependent cysteine proteases that play a role in various pathological processes, including neurodegeneration and angiogenesis. By inhibiting calpain activity, SK-1011 aims to suppress the formation of new, leaky blood vessels in the retina and protect retinal cells from damage. It has been evaluated in Phase 2 clinical trials in Japan, where it was administered at a dose of 60 mg twice daily.
