Overview

SK2109 is a long-acting implantable formulation of varenicline tartrate currently in preclinical development by Shenzhen Sciencare Pharmaceutical for the treatment of tobacco use disorder and smoking cessation. Varenicline is a small molecule that acts as a partial agonist at the α4β2 nicotinic acetylcholine receptors (nAChRs) in the brain. This mechanism provides a dual therapeutic effect: it partially stimulates the receptors to alleviate nicotine withdrawal symptoms and cravings, while also competitively inhibiting the binding of nicotine from cigarettes, thereby reducing the reinforcing rewards of smoking. By utilizing a long-acting implant delivery system, SK2109 aims to provide continuous therapeutic drug levels and improve patient adherence compared to traditional twice-daily oral varenicline regimens.
