SKB103 is an investigational antibody-drug conjugate (ADC) developed by Sichuan Kelun-Biotech Biopharmaceutical Co., Ltd., specifically targeting Claudin 18.2 (CLDN18.2). CLDN18.2 is a tight junction protein that is highly expressed in several types of solid tumors, including gastric, gastroesophageal junction, and pancreatic cancers, while having restricted expression in normal tissues, making it a promising therapeutic target. SKB103 utilizes Kelun-Biotech's proprietary ADC platform, likely employing a humanized monoclonal antibody conjugated to a potent cytotoxic payload, such as a topoisomerase I inhibitor. The drug is designed to selectively bind to CLDN18.2-expressing cancer cells, undergo internalization, and release its payload to induce cell death through DNA damage. It is currently undergoing Phase 1/2 clinical evaluation for patients with advanced solid tumors who have failed standard therapies.
