Overview
SKB264 + KL-A167 is a combination therapy of two investigational anticancer biologics used in clinical trials for various advanced malignancies. **SKB264** (also known as Sacituzumab tirumotecan or MK-2870) is an **antibody-drug conjugate** (ADC) targeting TROP2, designed with a belotecan-derived topoisomerase I inhibitor payload attached via a hydrolytically cleavable linker, enabling membrane-permeable payload release and bystander effect. **KL-A167** (Tagitanlimab) is a **humanized monoclonal antibody** targeting programmed death-ligand 1 (PD-L1), thus inhibiting the PD-1/PD-L1 immune checkpoint and restoring anti-tumor immunity. The combination is under phase II evaluation for treatment-naive advanced non-small cell lung cancer (NSCLC) and early-stage, high-risk ER+/HER2- breast cancer. Both drugs are developed by Sichuan Kelun-Biotech and Harbour BioMed, with SKB264 focused on targeted cytotoxic delivery and KL-A167 acting as an immune checkpoint inhibitor[3][5][6].
