Overview

skimmiarepin A is a naturally occurring protolimonoid (apotirucallane triterpenoid) isolated from the trunk bark of the Bael tree (*Aegle marmelos*) and other Rutaceae species. It acts as a potent inhibitor of hypoxia-inducible factor-1 (HIF-1) activation, with an IC50 of 0.063 µM in human breast tumor T47D cells. Mechanistically, skimmiarepin A suppresses mitochondrial respiration by selectively inhibiting complex I (NADH dehydrogenase) of the mitochondrial electron transport chain. This inhibition blocks the hypoxia-induced accumulation of nuclear HIF-1α protein and downregulates HIF-1 target genes such as GLUT-1 and VEGF. Additionally, it disrupts cellular signaling pathways controlling protein translation, including the phosphorylation of translation initiation factor eIF2α and elongation factor eEF2, thereby obstructing tumor cell proliferation. It has also been shown to exhibit moderate insecticidal activity.
