Skin-derived mesenchymal stem cells (S-MSCs) are multipotent stromal cells isolated from the dermis, offering a more accessible alternative to bone marrow-derived MSCs. These cells possess significant immunomodulatory and regenerative properties. In preclinical models of atherosclerosis, S-MSCs have been shown to reduce plaque size by migrating to the lesion and modulating macrophage phenotypes from pro-inflammatory to anti-inflammatory states. This effect is mediated through the NF-κB-dependent release of prostaglandin E2 (PGE2), which acts on EP2 and EP4 receptors to stimulate interleukin-10 (IL-10) production and suppress tumor necrosis factor-alpha (TNF-α). AOP Health is developing these cells (under the brand names AMESANAR and EBESANAR) for clinical applications, including the treatment of therapy-resistant chronic venous ulcers and epidermolysis bullosa.
