Overview

SKL2001 (frequently referred to in literature as SKL20001) is a synthetic small-molecule agonist of the canonical Wnt/β-catenin signaling pathway. It acts by directly binding to the Armadillo repeat domain (specifically repeats 3–5) of β-catenin, which competitively disrupts its interaction with the scaffold protein Axin. By preventing Axin from targeting β-catenin for phosphorylation and subsequent proteasomal degradation, SKL2001 stabilizes intracellular β-catenin, promoting its accumulation and nuclear translocation to activate downstream transcription factors. SKL2001 is widely utilized as a chemical probe in preclinical research to study mesenchymal stem cell differentiation, bone defect repair, oncology, and protective mechanisms against alcohol-induced liver injury.
