SKLB-V02 is a recombinant protein developed by the State Key Laboratory of Biotherapy at Sichuan University and West China Hospital for the treatment of fibrotic diseases, particularly systemic sclerosis (SSc) and its associated complications. It functions as a TGF-beta trap, incorporating the extracellular domain of the human TGF-beta receptor II (TβRII). By acting as a decoy receptor, SKLB-V02 binds with high affinity to and neutralizes the pro-fibrotic cytokines TGF-β1 and TGF-β3. This inhibition prevents the activation of the TGF-beta signaling pathway, which is a primary driver of myofibroblast differentiation and excessive extracellular matrix production in scleroderma and pulmonary fibrosis. The drug is currently undergoing Phase I clinical evaluation in China to assess its safety, tolerability, and preliminary efficacy in patients with diffuse systemic sclerosis and interstitial lung disease.
