SKM104-MA-L2 is a first-in-class topoisomerase I inhibitor-based antibody-drug conjugate (ADC) that targets GPCR1, a highly selective and abundant therapeutic target in pancreatic ductal adenocarcinoma (PDAC) and certain other cancers. This ADC consists of an anti-GPCR1 monoclonal antibody conjugated to an exatecan payload via a linker. The drug has shown robust preclinical anti-tumor activity and a high therapeutic index in models of pancreatic cancer, supporting its potential as a novel treatment approach where irinotecan-based regimens are standard. SKM104-MA-L2 is being developed by SkyMab Biotherapeutics, with current preclinical development focusing on oncology indications, particularly pancreatic cancer, pancreatic ductal adenocarcinoma, solid tumors, and triple negative breast cancer[1][4][7].
