Overview

The SKP2-CKS1 program is an AI-driven drug discovery initiative by Iktos focused on developing small-molecule inhibitors of the protein-protein interaction (PPI) between S-phase kinase-associated protein 2 (SKP2) and cyclin-dependent kinase regulatory subunit 1 (CKS1). This interaction is a critical component of the SCF-Skp2/Cks1 E3 ubiquitin ligase complex, which targets the tumor suppressor protein p27 for degradation. By disrupting this PPI, the inhibitors aim to stabilize p27 levels, leading to cell cycle arrest and inhibition of tumor growth in cancers dependent on the Skp2-p27 axis. Iktos utilizes its proprietary generative AI platform, Makya, and synthesis planning tool, Spaya, along with MTBench cellular technology to identify and optimize these novel PPI inhibitors.
