Overview

Skp2 siRNA is a research-grade small interfering RNA (siRNA) reagent used to silence the expression of the S-phase kinase-associated protein 2 (SKP2) gene. SKP2 is an F-box protein that serves as the substrate-recognition component of the SCF (Skp, Cullin, F-box) E3 ubiquitin ligase complex. It plays a pivotal role in cell cycle regulation by targeting tumor suppressors, such as p27Kip1, p21, and p57, for ubiquitin-mediated degradation. By utilizing the RNA interference (RNAi) pathway, Skp2 siRNA reduces SKP2 protein levels, leading to the stabilization of these tumor suppressors, which in turn induces cell cycle arrest and apoptosis. Preclinical studies have demonstrated its efficacy in inhibiting proliferation and reversing drug resistance in various malignancies, including multiple myeloma, acute myeloid leukemia, and prostate cancer. It is currently utilized as a laboratory tool and is not an approved therapeutic.
