Skullcapflavone II is a naturally occurring flavonoid compound primarily derived from the roots of *Scutellaria baicalensis*, a plant widely used in traditional herbal medicine. Chemically classified as a tetramethoxyflavone (C19H18O8), it features methoxy and hydroxy substitutions on its flavone backbone[2][5]. Skullcapflavone II exhibits multiple biological activities including anti-inflammatory and antimicrobial effects. It has demonstrated the ability to regulate osteoclast differentiation and function[1][7], inhibit cell proliferation by inducing G2/M phase arrest in hepatic stellate cells (suggesting potential for treating hepatic fibrosis)[6], and suppress production of pro-inflammatory cytokines relevant to atopic dermatitis[8]. Mechanistically, it acts as an inhibitor of DNA replication-related proteins (disrupting cell cycle progression), modulates TGF-beta/Smad signaling pathways, cyclooxygenase (COX) activity[3][7], and inhibits NAD(P)H dehydrogenase (quinone)[2]. Preclinical studies suggest therapeutic potential for allergic asthma[3][7], hepatic fibrosis[6], atopic dermatitis[8], and possibly other inflammatory or infectious conditions.
