Overview

SKWX301 is a human monoclonal antibody designed to block T-cell immunoglobulin and mucin domain-containing protein 4 (TIM-4), a scavenger receptor primarily expressed on macrophages and dendritic cells. TIM-4 is involved in the recognition and phagocytosis of apoptotic cells (efferocytosis) and has been shown to suppress anti-tumor immunity by promoting the degradation of ingested tumor antigens via the autophagy pathway. By blocking TIM-4, SKWX301 prevents macrophage-mediated phagocytosis of cancer cells and enhances antigen presentation, thereby boosting the anti-tumor immune response. Preclinical studies in murine colon carcinoma models have demonstrated that SKWX301, particularly when used in combination with anti-PD-1 checkpoint inhibitors, significantly inhibits tumor growth and improves overall survival. The antibody is engineered with an Fc fragment to increase its stability and pharmacokinetic profile compared to single-domain antibodies.
